Characterising the regulation of DNA repair activities and consequences of B-cell neoplasia associated oncogene expression in the germinal center.
 
Background
EU countries face large health challenges to combat chronic diseases including immune disorders. Recently, systems medicine has emerged as a promising discipline to accelerate the translation of basic research into applications for improved diagnostics and personalized treatment. Its power arises from the integration of laboratory and computational approaches crossing research disciplines and sectors to solve biomedical and clinical questions. The germinal center (GC) is a specialised anatomical compartment within secondary lymphoid tissues where lymphocytes interact and differentiate. Perturbations of the GC reaction contributes to autoimmunity and malignant transformation that underlies rheumatoid arthritis (RA) and B-cell lymphoma (BCL).
 
Approach
COSMIC will focus on the role of the GC in B-cell lymphoma (BCL) and rheumatoid arthritis (RA). Elucidating the cellular and molecular mechanisms of the GC reaction is esential to understand the ontogeny of RA and BCL. The PhD candidate will study the GC B-cell specific regulation of DNA repair fidelity and the effects of GC B-cell specific oncogene overexpression on GC resolution using genetically engineered human and mouse cell lines, transgenic mouse models, and 'multi-OMICS' approaches.
 
Our department
The Pathology department of the Academic Medical Center Amsterdam harbors several research groups that focus on elucidating the molecular mechanisms of B-cell lymphomagenesis and identifying novel therapeutic targets. Our department is part of the LYMMCARE (Lymphoma and Myeloma Center Amsterdam) knowledge center, which brings together biomedical and clinical expertise on B-cell malignancies, and allows for the rapid translation of novel scientific and clinical findings.
 
Your experience
  • Candidates should have a Master’s degree in Biology, Biomedical Sciences, Biochemistry, or similar with a strong background in immunology and molecular biology.
  • Experience with molecular biology and biochemistry of DNA repair.
  • Experience with laboratory animal husbandry and experimentation.
  • Basic statistical knowledge and affinity with OMICS approaches.
  • Excellent higher education track record and strong scientific curiosity. 
  • Fluent spoken and written English skills.
In addition, experience with the following would be helpful:
  • Experience in systems biology and systems medicine.
  • Experience in computational approaches and computer modelling.
